dc.description.abstractObjetivo: avaliar dos níveis de agregados de plaquetas-leucócitos circulantes em mulheres com câncer de mama. Métodos: entre 2018 e 2019, realizou-se um estudo corte transversal com 27 mulheres com diagnóstico de câncer de mama e idade entre 18 e 60 anos e 15 mulheres saudáveis (controles). Para a avaliação do agregado plaquetário circulante, foram coletadas amostras de sangue periférico no momento da realização dos exames laboratoriais de rotina para diagnóstico e antes do tratamento. A análise do agregado plaquetário foi realizado utilizando anticorpos monoclonais por citometria de fluxo. Testes de Mann-Whitney e Kruskal -Wallis foram usados para análise de medianas entre dois e três grupos, respectivamente. Foram considerados estatisticamente significativos valores de p < 0,05. As analises foram realizadas no graphpad v7.0. Resultados: Nas análises dos percentuais do agregados plaquetaslinfócitos (AGP-linfócitos), plaquetas-neutrófilos (AGP-neutrófilos) não foram observadas diferenças significativas entre as pacientes e os controles. No entanto, foi observado que as pacientes apresentaram valores percentuais elevados de agregado plaquetas-monócitos (AGP-monócitos) quando comparado aos controles (p<0,0001). Não foram observadas diferenças significativas nos níveis percentuais do AGPlinfócitos e AGP-neutrófilos entre os subtipos luminais A/B, HER2+ e triplo negativo; entre esses subtipos tumorais e controles. Os valores percentuais do AGP-monócitos foram elevados nos subtipos luminais A/B, HER2+ e triplo negativo quando comparado aos controles (p=0,008; p=0,0001; p=0.0002, respectivamente). No entanto, não foram observadas diferenças significativas percentuais do AGP-monócitos entre os subtipos tumorais. Conclusão: O presente estudo mostrou o envolvimento do AGP-monócito no câncer de mama triplo negativo e HER2+, sendo esses subtipos tumorais mais agressivos e pior prognostico. Com base nesses dados ficou clara a importância de novos estudos que tenham como base investigação do papel das interações entre as plaquetas e as células do sistema imune no câncer de mama. As moléculas envolvidas nas ligações entre plaquetas e leucócitos podem ser possíveis alvos terapêuticos.pt_BR

It was observed that patients had high percentages of platelet-monocyte aggregate (AGP-monocytes) when compared to controls (p<0.0001). No significant differences were observed in the percentages of AGP-lymphocytes and AGP-neutrophils between A / B, HER2 + and triple negative light subtypes; between these tumor subtypes and controls. The percentage values of AGP-monocytes were elevated in the A / B, HER2 + and triple negative light subtypes when compared to controls (p = 0.008; p = 0.0001; p = 0.0002, respectively). No significant percentage differences in AGP-monocytes were observed between tumor subtypes. Conclusion: The present study showed the involvement of monocyte AGP in triple negative breast cancer and HER2 +, being these tumor subtypes more aggressive and worse prognosis. Based on these data, the importance of further studies based on the role of interactions between platelets and immune system cells in breast cancer has become clear. Molecules involved in platelet-leukocyte binding may be possible therapeutic targets.pt_BR
